Can Somebody tell me how to use IntroVideo in Autoplay Media V4.0.0.3

That specific feature no longer exists as of 4.0. The best way I know of to get your project to run an intro video is to set up the first page with a video clip set to run automatically (Page Properties -> On Show -> play clip would be the most direct way). Then, if you are using the Media Player Object, set an On Stream End event to Page.Jump to your "second" page.
